High Performance Growth and Production
With the increasing demand for non-animal media for mammalian cell cultures, vegetable protein hydrolysates play a key role.
These vegetable protein hydrolysates have a proven function in:
- growth stimulation (amino acid replacement and replacement of serum components),
- cell integrity, and
- metabolite production
The amount of free amino acids and short peptides in a protein hydrolysate is important as they can be taken up directly and used by the cell as amino acid source. The stimulating effect of protein hydrolysates can also be the result of peptides functioning as a carrier for e.g. lipids, metal ions or toxic substances.
For several hybridoma cell lines it was shown in T-flask and spinner tests that vegetable protein hydrolysates can indeed replace serum when added in combination with 0.5-1% ITS (insulin, transferrin and selenium). Especially in spinner tests the growth of hybridoma cells and the production of product was highly stimulated when specific wheat gluten protein hydrolysates of the HyPep™ series were supplemented in a range of 1-4 g/L to standard DMEMF/F12 or IMDM media. Not only the growth of mammalian cells was stimulated but the cells also seemed to be better protected against shear stress.
